Inhibitory effect of sea horse Hippocampus kelloggi ethanol extract on antigen-stimulated degranulation in rat basophilic leukemia RBL-2H3 cells

Summary:Sea horse Hippocampus kelloggi is used as a traditional medicine in Asia for a tonic. In this study, we prepared a 35% ethanol extract of sea horse (SHEtEx.35), and then demonstrated the effects of SHEtEx.35 on antigen (Ag)-stimulated degranulation in rat basophilic leukemia RBL-2H3 cells. The inhibition level of Ag-stimulated degranulation by SHEtEx.35 gradually increased to 25,μg/mL, but the inhibition levels of Ag-stimulated degranulation by SHEtEx.35 did not change at concentrations above 25,μg/mL. In order to elucidate the underlying mechanisms, we examined the ešects of SHEtEx.35 on early intracellular signaling events (intracellular free Ca2+ ([Ca2]i) concentration, and intracellular reactive oxygen species (ROS) production) by antigen stimulation. Treatment with SHEtEx.35 markedly suppressed antigen-stimulated activation of PLCγ1 and elevation of [Ca2+]i. However, SHEtEx.35 did not suppress antigen-stimulated ROS production. These results indicate that inhibition of antigen-stimulated degranulation by SHEtEx.35 is mainly due to direct inhibition of PLCγ1 activation. Our findings suggest that SHEtEx.35 would be beneficial for alleviating symptoms of type I allergies. -

Summary:アサヒガニのゾエアから稚ガニまでの発育にともなう消化酵素活性の変化を実験室において調べた。4,000尾の幼生を水温26.3±1.6℃の0.5m3水槽で飼育した。ゾエア1~4齢まではアルテミア幼生で飼育し、その後、稚ガニまでは成長段階に併せて冷凍アサリ、アミ、コペポーダなどを給餌した。トリプシン様酵素およびアミラーゼの活性を孵化直後から孵化後54日まで測定した。両酵素はゾエア1齢から明らかに認められ、幼生の成長にともなって活性は上昇し、とくに孵化後16日からの活性の上昇は顕著であった。体重の著しい増加も孵化後16日頃から観察され、消化酵素活性の上昇とよく一致した。中腸腺の機能的な分化はゾエア5齢(孵化後16日)で明らかに見られ、この発育段階で消化系の基本的な構造が整ったことを示唆している。トリプシン様酵素およびアミラーゼの活性上昇は組織学的な観察により調べた中腸腺の分化と発達によく対応した。
